About Electron Microscopy, NeCEN, Leiden, The Netherlands

NeCEN is an open access facility for high resolution cryo-electron microscopy of biological samples. Two state-of-the-art Titan Krios transmission electron microscopes allow efficient and automated high resolution data collection in its two most broadly used forms, single particle analysis (SPA) and cryo-electron tomography (cryo-ET). These imaging techniques allow visualisation of biomolecular structures, such as proteins, macromolecular complexes, bacteria, and cell organelles at subnanometer resolution in close-to-native conditions. The facility is equipped with a BSL2 lab and microscope rooms.

 

Instruments Available:

The Talos L120C is a side entry 120KeV TEM. It allows for screening samples in negative stain and cryo conditions. It is used to optimize samples before they are imaged at the Krios. All necessary ancillaries equipment are available on-site for sample optimization.

Titan Krios 1 is a dedicated instrument for EFTEM (energy-filtered TEM) and cryo-tomography. This microscope has a SFEG source and is equipped with a Gatan GIF+K2 DED (integrated into EPU allowing auto data collection) and volta phase plate. It is an instrument for both tomography and automated single particle acquisition.

Titan 2 is a specialised instrument for single particle acquisition. This microscope has a XFEG source, a Cs corrector and a FEI Falcon 3 counting mode DED camera. It is a dedicated instrument for automated single particle acquisition.

User Guide

Titan Krios 1 is a dedicated instrument for energy-filtered TEM (EFTEM) and cryo-electron tomography. This microscope is equipped with a Gatan GIF+K2 DED as well as a volta phase plate. It is an instrument for both tomography and automated single particle acquisition.

Titan Krios 2 is a specialised instrument for single particle acquisition. This microscope has a Cs corrector, a Falcon 3 counting mode DED camera and volta phase plate.
