About Electron Microscopy, Oxford, UK

OPIC houses state-of-the-art biosafety containment laboratories at ACDP category 3 and DEFRA 4 levels of containment. This facilitates the study of a number of viruses that are important to human and animal health.  We have a range of state-of-the-art equipment for transmission electron microscopy of biological specimens, from small macromolecular complexes to cells. Our facilities are available to researchers from across the University of Oxford, and external users form academia and industry.

OPIC houses state-of-the-art equipment for cryo-EM. This includes a Titan Krios (Thermo Fisher Scientific) 300-kV transmission electron microscope equipped with Falcon III and K2 Summit/GIF (Gatan) direction electron detectors, in addition to a Glacios (Thermo Fisher Scientific) 200-kV, also equipped with a Falcon III, as well the facilities for the preperation of cryo-EM sample grids.

Autogrid clipping station for Glacios/Krios cryo grids

Leica GP2 plunge freezer.  This plunge freezer blots from one side only, but the sample can be rotated to blot both sides.  Located in Yellow suite of OPIC, so access will need to be authorised.

Glacios (Thermo Fisher Scientific) 200-kV equipped with a direct detector Falcon III, running EPU software.  Primarily used for screening of grids and preliminary data analysis. 

Titan Krios (Thermo Fisher Scientific) 300-kV transmission electron microscope equipped with Falcon III and K2 Summit/GIF (Gatan) direction electron detectors, capable of operating at ACDP category 3 and DEFRA 4 levels of containment. Gatan energy filtered BioQuantum967, volta-potential phase plate. Software includes EPU and serial EM. Optimised for electron tomography and single particle of small/flexible macromolecular complexes.

User Guide

If you already have grids prepared and prescreened, you can apply for Krios access for high resolution data acquisition (single particle or electron tomography). For initial grid screening and preliminary data analysis we would suggest applying for time on the Glacios.  If you require assistance during project work up, for example making, optimising and screening cryo-EM grids, please apply for access to the vitrobots or GP2, when we can assign the appropriate microscopes for the project stage.
