The workshop will focus on the analysis and optimisation of the sample quality of the purified macromolecules or macromolecular complexes. Attendees will learn that the quality of the sample is essential for the successful cryo-electron microscopy studies and see directly the effect of a poor sample quality.
The lectures will emphasize the importance of proper sample characterization and quality check for the success in following structural studies and give the theoretical background to the methods determining the sample suitability for Cryo-EM studies (SEC-MALS, light scattering, CD spectroscopy, MS, differential scanning fluorimetry and calorimetry, negative staining). Hands-on sessions will be aimed at obtaining practical skills with DLS (sample homogeneity and presence of aggregates), CD spectroscopy (structural intactness), DSC and nano-DSF (thermal stability) and performing the buffer screen to optimize sample conditions (pH, salt, additives).
The workshop will take place at the Masaryk University campus in Brno-Bohunice.
