Talk 1: Beyond static structural models: Analysis of continuous protein flexibility by cryoEM and its application to SARS-CoV-2

Speaker: José María Carazo and Marta Martinez

Abstract:  Our reprocessing of Cov-2 S protein in the prefusion state, in collaboration with McLellan and Tagare’s laboratories, has allowed us to address the whole range of spike flexibility in a direct manner. We have used Principal Component Analysis instead of the standard discrete class-oriented approach, concluding that movements are continuous and identifying the main axis of conformational variability.

 

Talk 2: An image processing pipeline for SARS-CoV-2 CryoEM: a single method is not enough

Abstract:   Results presented in the previous talk have required novel image processing workflows. We show in this seminar the approach we have used to fight structural artifacts reducing map bias. The corner stone is the use of “meta protocols”, looking for consensus among algorithms and implementations as different as possible in order to get reliable estimates of the parameters involved during structural elucidation.

 

Talk 3: Integrating structural information and biomedical annotations: 3DBionotes-Covid19 Portal

Abstract: Motivated by the emergence of the COVID-19 pandemic, a new section has been incorporated in 3DBionotes-WS, collecting and providing centralized access to available structural information on SARS-CoV-2 viral proteins as well as on other related viruses or interacting molecules. Considering the importance of cryo-EM-derived information for the understanding of SARS-CoV-2, we present an additional group of annotations to evaluate the quality of cryo-EM maps and the fit to their atomic models using different scores. In particular, the map quality is analysed by local resolution using DeepRes, while the fit and resolvability of the built atomic model can be studied with FSC-Q and Q-score.
